Ceruloplasminisamultifunctionalcopper-containingferroxidasepresentinserum.Ceruolpasministhemajortransporterofcopperintheblood,carryingmorethan95%ofthecopperfoundinplasma.Itusescoppertocoupletheoxidationofsubstratewiththereductionofoxygentowater.Ceruloplasmin-mediatedoxidationofferrous(Fe2+)irontoferric(Fe3+)ironcontributestotheregulationofironeffluxincellsthathavetransportableironstores.Inheritedloss-of-functionmutationsinceruloplasminhasbeenlinkedtoaceruloplasminemia.Ceruloplasminisknowntoformcomplexeswithcationicproteinsduringinflammationandisalsoinvolvedinpreventingiron-inducedoxidativedamage.IncreasedceruloplasminlevelshavebeenobservedinceliacandCrohn′spatients.Elevatedserumceruloplasminhasalsobeenlinkedtocancer,pregnancy,rheumatoidarthritis,schizophrenia,andAlzheimer′sdisease,whereasitslowlevelshavebeenlinkedtoWilson′sandMenkes′diseases.Hence,measurementofceruloplasminactivityinbiologicalsamplescanprovideusefulinsightsintoseveralmetabolicandpathologicalconditions.
TheCeruloplasminActivityColorimetricKitprovidesafastandeasy-to-useprocedureformeasuringceruloplasminactivityinserum.Ceruloplasminactivityisdeterminedusingsubstrateoxidation,whichresultsinacolorimetric(560nm)productproportionaltotheenzymaticactivitypresent.Oneunitofceruloplasministheamountofenzymethatwilloxidize1μmoleofsubstrateperminuteat25°C.
