      • ESR1 CISH Probe is designed for the qualitative detection of human ESR1 gene in formalin-fixed, paraffin-embedded specimens by chromogenic in situ hybridization (CISH).
      • Recommend Usage:
      • The product is ready-to-use. No reconstitution, mixing, or dilution is required. Bring probe to room temperature (18-25°C) and mix briefly before use.
      • Supplied Product:
      • Reagent Provided:1. Digoxigenin-labeled polynucleotides targeting sequences mapping in 6q25.1* (chr6:152,097,455-152,478,947) harboring the ESR1 gene2. Formamide based hybridization buffer*according to Human Genome Assembly GRCh37/hg19
      • Storage Instruction:
      • Store at 2-8°C in an upright position. Return to storage conditions immediately after use.
      • Note:
      • The probe is intended to be used in combination with the CISH Implementation Kit 1 (Catalog #: KA5365), which provides necessary reagents for specimen pretreatment and post-hybridization processing.Hybridization signals of digoxigenin-labeled polynucleotides appear as brown- to dark brown colored distinct dots (ESR1 gene region).Normal situation: In interphases of normal cells or cells without an amplification involving the ESR1 gene locus, two distinct dot-shaped brown signals per nucleus appear.Aberrant situation: In a cell with amplification of the ESR1 gene locus or aneusomy of chromosome 6, multiple copies of the brown signal or brown signal clusters will be observed. Other signal distribution may be observed in some abnormal samples which might result in a different signal pattern than described above, indicating variant rearrangements. Unexpected signal patterns should be further investigated.

      • This gene encodes an estrogen receptor, a ligand-activated transcription factor composed of several domains important for hormone binding, DNA binding, and activation of transcription. The protein localizes to the nucleus where it may form a homodimer or a heterodimer with estrogen receptor 2. Estrogen and its receptors are essential for sexual development and reproductive function, but also play a role in other tissues such as bone. Estrogen receptors are also involved in pathological processes including breast cancer, endometrial cancer, and osteoporosis.
