ADAM substrate is a FRET based fluorogenic substrate for ADAM metalloproteinases. The substrate contains the acceptor DABCYL, which acts as an acceptor for the donor, EDANS. Activity is quantified by release of fluorescent EDANS which excites at 365 nm and emits at 490 nm. ADAMs cleave extracellular portions of transmembrane proteins such as HER2 which is subsequently activated. Some ADAMS are also known as sheddases.
