HybridSPE-PhospholipidtechnologyisasimpleandgenericsampleprepplatformdesignedforthegrosslevelremovalofendogenousproteinandphospholipidinterferencesfromBIOLOGicalplasmaandserumpriortoLC-MSorLC-MS/MSanalysis.Biologicalplasmaorserumisfirstsubjectedtoproteinprecipitationviatheadditionandmixingofacidifiedacetonitrile.PrecipitatedproteinsarethenremovedbycentrifugationandtheresultingsupernatantisloadedontheHybridSPE-Phospholipid96-wellplateorcartridgewhichactsasachemicalfilterthatspecificallytargetstheremovalofendogenoussamplephospholipids.ThephospholipidretentionmechanismisbasedonahighlyselectiveLewisacid-baseinteractionbetweentheproprietaryzirconiaionsfunctionallybondedtotheHybridSPE-Phospholipidstationaryphaseandthephosphatemoietyconsistentwithallphospholipids.TheresultingeluentisreadyforimmediateLC-MSorLC-MS-MSanalysis.
